In modern composites there is a critical trade-off between strength and toughness. Natural structures provide blueprints to overcome this, however this approach introduces another trade-off between fine structural manipulation and manufacturing complex shapes in practical sizes and times. Here we show that robocasting can be used to build ceramic-based composite parts with a range of geometries, possessing microstructures unattainable by other production technologies. This is achieved by manipulating the rheology of ceramic pastes and the shear forces they experience during printing. To demonstrate the versatility of the approach we have fabricated highly mineralized composites with microscopic Bouligand structures that guide crack propagation and twisting in three dimensions, which we have followed using an original in-situ crack opening technique. In this way we can retain strength while enhancing toughness by using strategies taken from crustacean shells.	Oct 2017

This route offers significant advantages in terms of mild conditions, ease of use and electrode architecture over other routes to nanomaterials synthesis for device applications. The resulting 3D bicontinous nanowire network films exhibited a single diamond topology of symmetry Fd3m (Q227) which was verified by Small angle X-ray scattering (SAXS) and Transmission electron microscopy (TEM) and holds great promise for potential applications in optoelectronics, photovoltaics and thermoelectrics. In suitable microstructures, the displacement resolution is sub-voxel (a voxel is the three-dimensional equivalent of a pixel), and both elastic and plastic deformations can be studied. This paper reviews recent work in which three-dimensional in situ observations of deformation have provided unique insights that support both continuum and heterogeneous microstructure-dependent models of damage development in a range of materials. The examples presented include; crack propagation in a quasi-brittle porous material (polygranular graphite), sub-indentation radial and lateral cracking in a brittle polycrystalline ceramic (alumina); plastic deformation and damage development underneath indentations in a ductile metal (Al-SiC composite) and a ceramic matrix composite (SiC-SiCfibre). These examples show how material properties can be obtained by analysis of the displacement fields, how such measurements can be used to better define the applied loading on small test specimens and how crack opening magnitude and mode may be extracted also. Some new directions for research are outlined, including the combined use of diffraction and imaging techniques on synchrotron X-ray facilities to map both elastic and inelastic strains. Three-dimensional digital image correlation (digital volume correlation) is applied to study deformation beneath indentations, mapping the relative displacements between high-resolution synchrotron X-ray computed tomographs (0.9 ?m voxel size). Two classes of material are examined: ductile aluminium-silicon carbide composite (Al-SiC) and brittle alumina (Al2O3). The measured displacements for Hertzian indentation in Al-SiC are in good agreement with an elastic-plastic finite element simulation. In alumina, radial cracking is observed beneath a Vickers indentation and the crack opening displacements are measured, in situ under load, for the first time. Potential applications are discussed of this characterization technique, which does not require resolution of microstructural features.	Nov 2013

There is growing evidence that these additives are often occluded within the crystal lattice. This promises an elegant means of creating nanocomposites and tuning physical properties. Here we use the incorporation of sulfonated fluorescent dyes to gain new understanding of additive occlusion in calcite (CaCO3), and to link morphological changes to occlusion mechanisms. We demonstrate that these additives are incorporated within specific zones, as defined by the growth conditions, and show how occlusion can govern changes in crystal shape. Fluorescence spectroscopy and lifetime imaging microscopy also show that the dyes experience unique local environments within different zones. Our strategy is then extended to simultaneously incorporate mixtures of dyes, whose fluorescence cascade creates calcite nanoparticles that fluoresce white. This offers a simple strategy for generating biocompatible and stable fluorescent nanoparticles whose output can be tuned as required.	Nov 2016

In this paper a 3D-printed X-ray chamber, which allows for in situ exchange of buffer and in situ optical transmission spectroscopy, is presented. The chamber is made of cyclic olefin copolymers (COC), including COC X-ray windows providing ultra-low SAXS background. The design integrates a membrane insert for in situ dialysis of the 100 µl sample volume against a reservoir, which enables measurements of the same sample under multiple conditions using an in-house X-ray setup equipped with a 17.4 keV molybdenum source. The design's capabilities are demonstrated by measuring reversible structural changes in lipid and polymer systems as a function of salt concentration and pH. In the same chambers optical light transmission spectroscopy was carried out measuring the optical turbidity of the mesophases and local pH values using pH-responsive dyes. Microfluidic exchange and optical spectroscopy combined with in situ X-ray scattering enables vast applications for the study of responsive materials.	Jul 2022

Since the COVID-19 outbreak, new structural data from many viral proteins have been provided at a very fast pace. This effort includes many cryogenic Electron Microscopy (cryo-EM) studies, together with more traditional ones (X-rays, NMR), using several modeling approaches and complemented with structural predictions. At the same time, a plethora of new genomics and interactomics information (including fragment screening and structure-based virtual screening efforts) have been made available from different servers. In this context we have developed 3DBionotes-COVID-19 as an answer to: (1) The need to explore multi-omics data in a unified context with a special focus on structural information and (2) the drive to incorporate quality measurements, especially in the form of advanced validation metrics for cryogenic Electron Microscopy. Withers Solid Earth 7, 1059 - 1073 DOI: 10.5194/se-7-1059-2016 Diamond Proposal Number(s): [10500] Open Access Show Abstract ▼ Abstract: A variable volume flow cell has been integrated with state-of-the-art ultra-high-speed synchrotron X-ray tomography imaging. The combination allows the first real-time (sub-second) capture of dynamic pore (micron)-scale fluid transport processes in 4-D (3-D + time). With 3-D data volumes acquired at up to 20 Hz, we perform in situ experiments that capture high-frequency pore-scale dynamics in 5–25 mm diameter samples with voxel (3-D equivalent of a pixel) resolutions of 2.5 to 3.8 µm. The data are free from motion artefacts and can be spatially registered or collected in the same orientation, making them suitable for detailed quantitative analysis of the dynamic fluid distribution pathways and processes. The methods presented here are capable of capturing a wide range of high-frequency nonequilibrium pore-scale processes including wetting, dilution, mixing, and reaction phenomena, without sacrificing significant spatial resolution. As well as fast streaming (continuous acquisition) at 20 Hz, they also allow larger-scale and longer-term experimental runs to be sampled intermittently at lower frequency (time-lapse imaging), benefiting from fast image acquisition rates to prevent motion blur in highly dynamic systems. This marks a major technical breakthrough for quantification of high-frequency pore-scale processes: processes that are critical for developing and validating more accurate multiscale flow models through spatially and temporally heterogeneous pore networks. Specific Pro-rich sequences with the consensus core motif PxxP adopt the PPII helix conformation upon binding to SH3 domains. For short Pro-rich peptides, little or no ordered secondary structure is usually observed before binding interactions. The association of a Pro-rich peptide with the SH3 domain involves unfavorable binding entropy due to the loss of rotational freedom on forming the PPII helix. With the aim of stabilizing the PPII helix conformation in the Pro-rich HPK1 decapeptide PPPLPPKPKF (P2), a series of P2 analogues was prepared, in which specific Pro positions were alternatively occupied by 4(S)- or 4(R)-4-fluoro-L-proline. The interactions of these peptides with the SH3 domain of the HPK1-binding partner HS1 were quantitatively analyzed by the NILIA-CD approach. A CD thermal analysis of the P2 analogues was performed to assess their propensity to adopt the PPII helix conformation. Contrary to our expectations, the Kd values of the analogues were lower than that of the parent peptide P2. These results clearly show that the induction of a stable PPII helix conformation in short Pro-rich peptides is not sufficient to increase their affinity toward the SH3 domain and that the effect of 4-fluoroproline strongly depends on the position of this residue in the sequence and the chirality of the substituent in the pyrrolidine ring.
